Abstract:
Sul1 and Sul 2 are key sulfate transporters in S. Cerevisiae which belong to the SLC26/SULP family. The SLC26/SULP family are one of the widely researched anion transporters and the previous research on Sul1 and Sul2 homologues served as the foundation for rational investigation of sulfate transport function. Two control strains were studied namely HK14 with an empty gene (∆sul1 ∆sul2) and HK14 with Sul1 gene (∆sul2). These strains were grown in SD agar plates without uracil and sulfur. The growth curves for these strains were obtained in sulfur starved media and SD liquid media without uracil and their absorbance were recorded at 600nm for a time period of 24 hrs. Four different concentration of ammonium sulfate (0mM, 2mM, 8mM, 38mM) was added to each strain to observe how this affect the growth of these organism. According to theory, S. Cerevisiae with the Sul1 gene and 38 mM concentration of ammonium sulfate should show the highest growth and S. Cerevisiae with an empty vector and 0mM ammonium sulfate should show the least growth. Two point mutated Sul1 gene strains were analyzed to determine if the point mutation helps in improving the function of Sul1 and improving the metabolism of the yeast.
